Standardized Tongkat Ali Root Extract

Tongkat Ali aka Long Jack (Eurycoma Longifolia)

Tongkat Ali also known as Eurycoma longifolia and is called Longjack in America, is an evergreen small tree that is known to produce a wide range of positive effects, especially to men. The herb typically grows in countries within South-East Asia such as Malaysia, Thailand and Indonesia. It is mainly due to the presence of eurycomanone in Tongkat Ali (a type of quassinoid) that makes it a marker for the identification of this plant and the level of Tongkat Ali in a product.

In the past, the roots of the Tongkat Ali tree is usually the part that is ground into powder, but direct consuming provides little or no benefits plus plant cellulose is indigestible. Therefore, technology to extract the active compounds by hot water has been used to obtain the water soluble active ingredients ie. Eurycomanone, polysaccharides, proteins and glycosaponin from the herb. Subsequently, spray drying has been employed to produce Tongkat Ali extract powder which can be made used for various applications such as additives and supplements. Because the expensiveness and preciousness of the herb, studies have been moving towards the direction of increasing the amount of active compounds extracted and the yield of final product.

Yellow tongkat ali is chosen because its the only tongkat ali species which has eurycomanone

Tongkat Ali (Eurycoma Longifolia): Everything You Need to Know

Tongkat ali is a traditional Asian remedy for male sexual performance. Advocates of traditional medicine also use this plant to help relieve other ailments and health issues including fevers, erectile dysfunction, and bacterial infections.

Studies suggest that tongkat ali may boost male fertility, relieve stress, and improve body composition, but research in these areas is limited.

People should be cautious about trying it if they have certain health conditions or are taking specific medications even though there has been promising research into tongkat ali’s properties.

Keep reading to learn more about tongkat ali’s traditional uses and what the scientific evidence says about its potential benefits and side effects.

What is Tongkat Ali?

Tongkat ali, or longjack, is an herbal supplement that comes from the roots of the green shrub tree Eurycoma Longifolia, which is native to Southeast Asian countries such as Malaysia, Indonesia, and Vietnam. Some species of the plant also grow in regions of Cambodia, Myanmar, and Thailand.

Tongkat ali literally means “Ali’s walking stick,” which makes reference to its aphrodisiac effects. Some people say that the “stick” refers to the plant’s long, twisted roots that some groups harvest for their medicinal value.

Other common names for E. longifolia are Long Jack, Malaysian ginseng, and Ali’s Umbrella.

The health benefits of tongkat ali likely stem from various compounds found in the plant.

Tongkat ali contains flavonoids, alkaloids, and other compounds that act as antioxidants. The benefits of Antioxidants is that it’s a compound that fights cellular damage caused by molecules called free radicals.

Usage of Tongkat Ali

1) Traditional use

Tongkat ali treats a range of ailments including postpartum depression, malaria, high blood pressure, and fatigue. Furthermore, Decoctions of tongkat ali roots have been used for centuries in Malaysia and Southeast Asia as an aphrodisiac for loss of sexual desire and impotence.

Tongkat ali has been referred to as Malaysia’s “home-grown Viagra” [4] with the Malaysian government investing considerable effort to license, develop, and sustain research into the potential health benefits of Eurycoma longifolia through a variety of governmental organizations, including the Forest Research Institute of Malaysia (FRIM).

2) Modern extracts

Numerous commercial tongkat ali supplements claim “extract ratios” from 1:20 to 1:200 without any information about bioactive constituents, extraction methodology (e.g. ethanol versus water), or extract purity.

Alcohol extracts of eurycoma have been studied in mice for antimalarial effects of concentrated eurycomalactone but also exhibit toxic effects at high doses (LD50 at 2.6 g/kg), which would preclude safe use in humans as a long-term dietary supplement . In contrast, hot-water root extracts standardized for known bioactive components (1% eurycomanone, 22% protein, 30% polysaccharides, 35% glycosaponin) have been demonstrated to be extremely safe at high doses and for long-term consumption.

Properly standardized hot-water extracts have a distinctly bitter taste due to the presence of quassinoids, which are recognized as some of the bitterest compounds in nature. Still, some tongkat ali extracts doesn’t taste bitter because either they’re not true Eurycoma longifolia root (there are many commercial examples of “fake” tongkat ali extracts) or are sub-potent in terms of bioactive constituents, and thus would also be expected to have low efficacy.

Because of tongkat ali’s reputation for libido benefits, there are several examples of dietary supplements labeled as Eurycoma longifolia, but containing none of the actual root, and instead being “spiked” with prescription erectile dysfunction drugs including tadalafil/Cialis, sildenafil/Viagra, and vardenafil/Levitra.

3) Laboratory and animal research

Bhat and Karim conducted an ethnobotanical and pharmacological review on tongkat ali, noting that laboratory research such as cell assay studies offer possible mechanistic support for the myriad traditional uses of tongkat ali, including aphrodisiac, antimalarial, antimicrobial, anti-cancer, and anti-diabetic effects.

Eurycoma’s anxiolytic effects have been demonstrated in a variety of behavioral tests, including elevated plus-maze, open field, and anti-fighting, suggesting an equivalent anti-anxiety effect to diazepam as a positive control. This is because Numerous rodent studies exist demonstrating reduced anxiety and improved sexual performance following tongkat ali feeding, with such effects thought to be due to a restoration of normal testosterone levels.

Animal studies have shown that many of the effects of the extract are mediated by its glycoprotein components. The mechanism of action of the bioactive complex polypeptides (“eurypeptides” with 36 amino acids) has been shown to activate the CYP17 enzyme (17 alpha-hydroxylase and 17,20 lyase) to enhance the metabolism of pregnenolone and progesterone to yield more DHEA (dehydroepiandrosterone) and androstenedione, respectively. This glycoprotein water-soluble extract of Eurycoma longifolia has been shown to deliver anti-aging and anti-stress benefits subsequent to its testosterone-balancing effects.

Oral toxicity studies (Wistar rats) have determined the LD50 of tongkat ali root extract as 2,000 mg/kg body weight (acute) and the NOAEL (no observed adverse effect level) as greater than 1,000 mg/kg body weight (28-day sub-acute feeding), resulting in a classification as Category 5 (extremely safe) according to the United Nations Globally Harmonized System of Classification and Labeling of Chemicals (GHS).

There are no reported adverse side effects in human studies of tali supplementation in addition to the very high safety profile demonstrated in the rodent toxicity studies. For example, one 2-month human supplementation trial [27] of twenty healthy males (age range 38–58), found high doses of Eurycoma longifolia extract (600 mg/day) to have no influence on blood profiles (hemoglobin, RBC, WBC, etc.) or any deleterious effects on measures of liver or renal function.

The typical dosage recommendations is based on traditional use and on the available scientific evidence in humans, including dieters and athletes, call for 50-200 mg/day of a water-extracted tongkat ali root standardized to 22% eurypeptides.

4) Human supplementation trials

Based on a long history of traditional use and confirmation of biological activity via cell culture and animal feeding studies, several human supplementation studies have been conducted to evaluate the potential benefits of tongkat ali for sexual function, exercise performance, weight loss, and vigor (mental/physical energy).

Importantly, all of the human trials have used the same water-extracted and standardized eurycoma root for which a patent has been issued jointly to the Government of Malaysia and the Massachusetts Institute of Technology (United States Patent #7,132,117).

The patent discloses a process whereby Eurycoma longifolia roots undergo an aqueous extraction combined with HPLC and size-exclusion chromatography to yield a bioactive peptide fraction (a 4300 dalton glycopeptide with 36 amino acids) that is responsible for its effects in maintaining testosterone levels.

Overall, the bioactive fraction of Eurycoma longifolia root delivers a demonstrated ability to improve testosterone levels, increase muscle size and strength, improve overall well-being, accelerate recovery from exercise, enhance weight loss, reduce stress, and reduce symptoms of fatigue.

NCBI(National Center for Biotechnology) undertook a study of the effects of tongkat ali root extract supplementation in moderately stressed subjects based on it’s long history of traditional medicinal use as an “anti-aging” remedy and the series of animal and human supplementation studies investigating its use as a physical and mental performance aid.

Our hypothesis was that tongkat ali supplementation may influence anabolic/catabolic stress hormone balance and mood state parameters in a group of volunteers with moderate stress levels.

Methods by NCBI to study Tongkat Ali

All procedures, measurements, and informed consent processes were reviewed and approved by an external third-party review board from NCBI(National Center for Biotechnology).

Subjects were recruited in and around Salt Lake City, Utah via flyers asking for volunteers with “moderate stress levels”. They screened approximately 75 subjects for moderate levels of psychological stress. Their intention was to complete the study with 60 subjects (30 subjects per treatment group). They used a screening survey that they themselves have used in past studies of stress/mood to identify individuals with moderately elevated levels of perceived stress. Subjects scoring 6 or greater on this screening survey indicated eligibility for enrollment into the supplementation study (a score of 6–10 indicates moderate stress).

Sixty-four (64) subjects (32 men and 32 women) were randomized to receive tongkat ali (TA; 200 mg/day of Physta™, Biotropics Malaysia Berhad; 32 subjects) or look-alike placebo (PL; 32 subjects) for 4 weeks. The 4-week duration was selected as more representative of persistent changes in mood state that may result from superior hormone balance, as opposed to short-term changes in emotions that may be more closely linked with stressors of daily living.

At Baseline (week 0) and Post-supplementation (week 4), they assessed Mood State and Hormone Profile as their primary outcome measurements. Secondary measurements were made of liver enzymes (ALT; alanine aminotransferase and AST; aspartate aminotransferase; Alere Cholestech, Waltham, MA), body weight, and body fat percentage (Tanita; TBF-300A, Arlington Heights, IL).

Mood State (Vigor, Depression, Anger, Confusion, Fatigue, and Anxiety) was assessed using the validated Profile of Mood States (POMS) survey. Hormone profile (cortisol and testosterone) was assessed in saliva samples collected at three time points during each collection day (morning, afternoon, and evening). Saliva samples were analyzed for free cortisol and free testosterone by enzyme immunoassay (Salimetrics; State College, PA).

Results were analyzed by one-way analysis of variance (ANOVA) with significance set at p < 0.05.

Sixty-three subjects (32 men and 31 women) completed the study, with one woman in the supplement group lost to follow up (did not return final samples).

Potential health benefits

Some studies suggest that it may help treat male infertility, improve mood, and increase muscle mass even though most of the alleged health benefits of tongkat ali are still not well researched.

May increase testosterone levels and improve male fertility

Fortunately, Tongkat ali’s potential to increase testosterone in men with low levels of this primary sex hormone is well known and well documented which is a great news for men that have this problem.

Low testosterone can result from aging, chemotherapy, radiation treatments, some medications, injury or infection of the testicles, and certain diseases, such as chronic alcoholism and obstructive sleep apnea.

Low libido, erectile dysfunction, and in some cases, infertility are some of the effects of inadequate testosterone levels. However, it could treat these issues Since compounds in tongkat ali may boost low testosterone.

According to a 2016 review, animal and human studies suggest that the beneficial effects may include:

increased semen volumes, sperm count, sperm viability, and sperm motility
increased production of testosterone
enhanced erectile function and sexual performance

Also, taking 200 mg of tongkat ali extract per day significantly increased levels of this hormone to normal values in over 90% of participants according to a 1-month study in 76 older men with low testosterone.

What’s more, studies in both animals and humans show that taking tongkat ali stimulates sexual arousal and may improve erectile dysfunction in men.

Finally, tongkat ali may improve sperm motility and concentration, boosting male fertility.

Furthermore, taking 200 mg of tongkat ali extract per day significantly improved sperm concentration and motility after 3 months. The treatment helped over 14% of couples become pregnant according to one study in 75 male partners of couples with infertility.

Similarly, a 12-week study in 108 men ages 30–55 observed that taking 300 mg of tongkat ali extract daily increased sperm volume and motility by an average of 18% and 44%, respectively.

According to these studies, tongkat ali effectively treats low testosterone and infertility in some men, but more extensive research is needed.

May relieve stress

Tongkat ali may lower stress hormones in your body, decrease anxiety, and improve mood.

Tongkat ali are able to treat mood issues and found that tongkat ali extract was comparable to a common anti-anxiety medication in reducing symptoms of anxiety in mice according to a 1999 study.

Another study in the Journal of the International Society of Sports NutritionTrusted Source assessed stress hormones in 63 people taking tongkat ali extract or a placebo for 4 weeks.

The researchers saw significant improvements in anger, tension, and confusion in people taking the extract. Testosterone levels increased, and levels of the stress hormone cortisol decreased.

They concluded that tongkat ali might be an effective remedy for modern-day chronic stress, sleep deprivation, and exercise training.

Another study from 2018 looked at the combined effects of E. longifolia and multivitamins in moderately stressed but otherwise healthy participants. The participants reported enhanced vigor and emotional well-being. Glucose concentrations also decreased, which may also have contributed to the participants’ well-being.

May improve body composition

Tongkat ali is often claimed to boost athletic performance and increase muscle mass.

Also, tongkat ali may help your body use energy more efficiently, reduce fatigue, and improve endurance as it contains compounds called quassinoids, including eurycomaoside, eurycolactone, and eurycomanone.

This means that the supplement may act as an ergogenic aid, which is a substance that can enhance physical performance and improve body composition.

Those who took 100 mg of tongkat ali extract per day experienced significantly greater increases in lean body mass than those taking a placebo according to a small, 5-week study in 14 men participating in a strength training program.

They also lost more fat than participants in the placebo group.

What’s more, a 5-week study in 25 active older adults discovered that supplementing with 400 mg of tongkat ali extract daily significantly increased muscular strength, compared to a placebo.

However, consuming a drink with tongkat ali during exercise did not improve performance or strength any more than plain water according to an observation from a small study in cyclists.

These conflicting results suggest that tongkat ali may exhibit some ergogenic effects, depending on the dose and length of treatment, but more research is needed.

Studies show that tongkat ali may boost testosterone levels and help treat infertility in men, relieve stress, and possibly increase muscle mass. Still, more extensive research is needed.

Bone health

A 2018 review examined the evidence for E. longifolia on osteoporosis in rats. It concluded that E. longifolia seemed less effective than other traditional Malaysian herbs.

However, there could still be potential for its use in preventing bone loss, according to the review.

Cancer

Laboratory studies in test tubes indicate that the compounds are toxic to lung and cervical cancer cells as scientists are discovering and identifying new quassinoids in E. longifolia that may help fight cancer.

A review from 2018 identified 16 compounds in E. longifolia, isolated from various parts of the plant, that showed promising anticancer properties.

Another laboratory study indicated that quassinoids from E. longifolia were effective against human prostate cancer cells. However, the Endocrine Society contraindicates testosterone treatment in prostate cancer, so using the plant for this condition is not advisable.

Anti-inflammatory and antimicrobial effects

Some researchTrusted Source indicates that E. longifolia is effective against bacteria, including Escherichia coli and Salmonella typhi.
In test tube studies, extracts of the plant showed anti-inflammatory and antioxidant properties.

Possible side effects and dosage

The few studies on the use of tongkat ali in humans have not reported any side effects.

One study noted that taking 300 mg of tongkat ali extract daily was as safe as taking a placebo.

According to a 2016 review on the safety and toxicity of E. longifolia, it’s reported that it does not seem to have detrimental effects on sperm in test tubes when the scientists used it in therapeutic doses. However, animal studies show that in higher concentrations, it may be toxic.

Moreover, other studies suggest that taking up to 1.2 grams of tongkat ali extract per day is safe for adults, but this amount has not been used in research. Plus, no studies examine its long-term use, making it unclear whether the supplement is safe over longer periods.

Scientists still consider tongkat ali(E. longifolia) safe as long as people do not take it in high doses. The authors recommend taking 200–400 milligramsTrusted Source daily with caution, especially if the person is an older adult.

Still one study shows that the mercury content of 100 tongkat ali supplements from Malaysia found that 26% had levels of mercury higher than the recommended limit.

Although laboratory studies have indicated beneficial effects, these effects may not be the same in the human body as people who have hormonal cancers should be cautious of taking E. longifolia, as it may increase testosterone levels.

If you’re taking medications to lower their blood glucose, it’s wise to speak to your doctor first before taking E. longifolia, as it may increase the effects of these medications.

Furthermore, the effects of tongkat ali in children or pregnant and breastfeeding women have not been researched. Therefore, it’s unknown whether the remedy is safe for these populations.

Should you take tongkat ali?

Some studies suggest that tongkat ali may reduce anxiety and improve body composition, but research is limited.

It may also treat low testosterone, poor libido, and male infertility.

Research on the tongkat ali is still limited, and available studies only focus on short-term use. Still, tongkat ali does not appear to have adverse effects in doses up to 400 mg per day.

It’s unclear whether taking the supplements over longer periods is beneficial and safe.

Consult your healthcare provider to ensure proper safety if you’re interested in taking tongkat ali.
Additionally, keep in mind that some supplements may be contaminated with mercury. Plus, they’re not well regulated and may contain more or less tongkat ali than listed on the label. This is why it’s very important Look for a reputable brand that has been tested by a third party.

Lastly, due to the lack of research on the effects of tongkat ali to pregnant and breastfeeding women, It’s better for pregnant and breastfeeding women to never take tongkat ali at all. Additionally, those with medical conditions or taking medications should speak to their healthcare provider before taking tongkat ali.

Summary

Tongkat ali seems to be a promising remedy for several health issues. Some research suggests that it is beneficial for male fertility, sexual performance, and stress. It might also be an effective ergogenic aid.

Research also suggests that people with certain cancers should avoid using it. However, Some laboratory studies indicate the effectiveness of E. longifolia against cancer in test tubes.

Therefore, a person should check with their doctor before taking any herbal supplements as there are some safety issues for people who have certain health conditions and those who are taking specific medications.

Frequently Ask Questions:

What Is Eurycomanone Standardized Tongkat Ali Root Extract?
There are two forms of tongkat ali root extract available on the market today, traditional and standardized, traditional extract is strength rated by the amount of root used to produce a single gram of extract, this method of production is highly inefficient, allowing for a large margin of error in actual finished extract strength. This is because the active ingredients in a tongkat ali root extract will vary depending on the age and quality of the root used to produce the extract, rather than simply the amount of root used. An example of this would be as follows: If two separate batches of tongkat ali root extract were produced each using 50 grams of root to produce 1 gram of finished extract (1:50), the first batch using eurycomanone rich root while the second used roots with a low eurycomanone content, the two finished extracts would be completely different in strength, although each batch used 50 grams of root per 1 gram of finished extract.

How Is Standardized Tongkat Ali Root Extract Produced?
Standardized tongkat Ali root extract is produced in much the same way as traditional tongkat ali root extract, but rather than basing the strength rating on the amount of root used regardless of quality, we engage FRIM (Forest Research Institute of Malaysia) as Thrid Party Verification, who use analytic equipment in the form of ultra-high-pressure liquid chromatography tandem mass spectrometry to monitor the levels of several key active ingredients, primarily the eurycomanone content of the extract.

What Are The Benefits Of Standardized Extract?
Besides the obvious benefits of safety and quality through consistency, using analytic equipment to produce plant extracts stops the unnecessary waste of raw materials by adjusting the formula at a granular level. Additionally this method of production helps to reduce fraudulent activity in the supply chain, as the process is completely verifiable from start to finish.

Does Tongkat Ali really work?
Tongkat Ali works on several mechanisms involved with hormonal function and Intimacy performance. In other words, Tongkat Ali minimish ageing on the men reproductive system by increasing body’s free testo bio-availability.

  1. Stimulating the Leydig cells of your testicles to produce testo.
  2. Reducing the conversion of Testo to Estrogen
  3. Decrease in hormone binding globulin
  4. Reduce stress hormone

Estrogen levels of the average 54-year-old man are often higher than those of the average 59-year-old woman. Too much estrogen contributes to a wide range of health, weight gain and libido problems. High serum levels of estrogen also trick the brain into thinking that enough testo is being produced, thereby slowing the natural production of testo. Results of studies have confirmed Tongkat Ali may be of assistance in the following areas:

  • Create free testo level
  • Improving erectile strength without side effects
  • Increase in men fertility, sperm quality, motility and quantity
  • Natural energy booster
  • Improving strength during exercise with an anabolic effect on muscle growth
  • Decrease in hormone binding globulin (SHBG)
  • May promote weight loss

Can I take Tongkat Ali if I have high blood pressure?
Tongkat Ali will not aggravate high blood pressure as it has a mild anti-hypertensive vasodilatory effect. One of the reasons Tongkat Ali is beneficial for erectile dysfunction is because it assists vasodilation of penile circulation as confirmed in scientific research.

What recommended dosage for Tongkat Ali Root Extract?
Tongkat Ali dosage increases with age and is also based on the underlying hormonal deficiency. As such, the dosage of a 65 years old will need to be increased from that of a 35 year old. Taking this into consideration you may experiment to find the dosage that feels right for you.

Does Tongkat Ali have any side effects?
Our Tongkat Ali products, taken at their recommended dosage are very well tolerated and free of toxicity. One may experience an increase in metabolic body heat (thermogenesis) after taking Tongkat Ali and it is recommended to increase water intake. For individuals with high blood pressure and or diabetes then our Rainforest Herbs Tongkat Ali Plus Capsules formulation has been specially prepared to support male hormonal and cardiovascular health and antioxidant protection from free radical damage. In my experience each individual reacts slightly differently, so the product dosage that works for you may be different for another. High dosages may give more dramatic effects on libido within a shorter time frame of around 1-2 days, while lower dosages may take 1 week or more to notice the effect.

What is Red and Black Tongkat Ali?
Yellow, Red and Black Tongkat Ali are all unrelated medicinal plants that share a similar common name in some regions of Malaysia. However, the name Tongkat Ali is most commonly in reference to yellow Tongkat Ali (yellow) or Eurycoma longifolia, of the family:Simaroubaceae.

Can Tongkat Ali be taken by women?
Yes, Women in Borneo take Tongkat Ali as Energy Booster. Tongkat Ali is suitable in menopausal women as it helps maintain progesterone production from the ovaries and adrenal glands.

Is Tongkat Ali high in Heavy Metal?
Tongkat Ali are harvested in rain forest, thus the heavy metal have negligible levels. All root herb or vegetables must be thoroughly cleansed of surface soil after collection. All batches of health products manufactured under GMP & HACCP standards must undergo a heavy metal test (Atomic Absorption Spectrometry) to determine whether lead and other heavy metals are present.

How to maximize the effects of Tongkat Ali ?
Reduce Carbohydrate Intake and Regular exercise / workouts can improve the results and accelerate muscle growth and increase testo production and enhance libido.

What Is Eurycomanone And Why Is It So Important?
Eurycomanone is the active compound[1]found in the root of the eurycoma longifolia plant, more widely known as tongkat ali.[2]. Studies have shown that eurycomanone increases the amount of free testo available in the bloodstream by inhibiting the conversion of testo to estrogen, this causes an increase of testo to build up over a period of time. At higher concentrations, eurycomanone also acts as a phosphodiesterase type 5 inhibitor and is currently being studied for the treatment of erectile dysfunction.[3]

What Strength of Eurycomanone Standardized Tongkat Ali Root Extract is Best?
Eurycomanone is available in pure form, but this is not safe to consume and certainly would not be considered a tongkat ali root extract for therapeutic use. For safe human consumption standardized tongkat ali root extract starts at 0.5% eurycomanone and the upper limits are around 2.7% eurycomanone, in these strengths standardized tongkat ali root extract is used as a safe testo booster with the higher strength extracts also acting as a PDE5 inhibitor..[4]
